Determination of phenol - sulfuric acid method
1 Scope
This standard specifies the export of plant-derived foods polysaccharides content colorimetric assay.
This standard applies to the determination of the content of edible fungus polysaccharides, Chinese wolfberry, grapes, jujube, fruit juice and other plant-derived foods.
This standard does not add starch, dextrin food components.
The detection limit of the standard method of 0.5mg/kg.

The following terms and definitions apply to this document.

Polysaccharide crudepolysaccharides
Β-D- glucan with α-D- glucan or other carbon sugar or a series of main chain polymer compound. Molecular formula (C6H12O6) n.
4 Method summary
Polysaccharides under the action of concentrated sulfuric acid hydrolysis to monosaccharides, uronic and rapid dehydration derivative, is reacted with a phenol to generate orange solution,
Characteristic absorption at 490nm, a quantitative comparison with the standard series.
5 Reagents and materials
Unless otherwise specified, use only analytical reagents, test water should be consistent with the three water GB/T 6682-2008 stipulated.
5.1 sulfate (H2SO4), ρ = 1.84g/mL.
5.2 absolute ethanol (C2H6O).
5.3 Phenol (C6H6O), redistilled.
5.4 80% ethanol solution.
5.5 glucose (C6H12O6), before use at 105 ℃ temperature drying to constant weight.
5.6 80% phenol solution. Weigh 80g of phenol (5.3) in the 100mL beaker, dissolved in water, go to the 100mL flask set brown
