1 Scope
This Standard specifies the general principles, basic requirements, evaluation
methods and evaluation report expressions of efficacy evaluation for removing
of heavy metal by biological products.
This Standard applies to the effect evaluation for removing of heavy metals from
water by biological products.

3 Terms and definitions
The following terms and definitions are applicable to this document.
3.1
Biological products
Products which are made by biotechnology.
Note: Biological products in this Standard specifically refer to microorganisms
or enzyme products that have the function of removing heavy metals.
4 General principles
4.1 Evaluation can only be carried out when the label or manual of the biological
product has a clear name of the microorganism or enzyme product.
4.2 Scientifically and fairly carry out efficacy evaluation work; the evaluation
process and evaluation method are scientific.
4.3 Use statistical methods for comparison; no forgery or alteration of inspection
reports is allowed.
5 Basic requirements
5.1 In accordance with the requirements of relevant laws, regulations,
standards and product manuals, receive, store, hand over, prepare, recycle,
and return/destroy samples; formulate corresponding management systems
and procedures.
5.2 The laboratory that conducts the evaluation of microbial products shall meet
the grade-1 requirements of bio-safety.
5.3 The type, quantity, performance, range and accuracy of the instruments and
equipment used in the evaluation shall be able to meet the needs of the efficacy
evaluation. The utensils and materials required for the efficacy evaluation of
microbial products shall be treated aseptically.
5.4 The reagents used in the evaluation shall be analytical reagents; the water
shall conform to the grade-II water that is specified in GB/T 6682.
5.5 The heavy metals used in the evaluation are made of heavy metal standards
whose purity is greater than or equal to 98%, or standard materials which are
certified by the country and awarded with a standard material certificate, and
formulated into a standard solution of a certain mass concentration. The
medium shall be selected according to the type of microorganisms.
5.6 After the evaluation experiment is over, the experimental materials shall be
treated in a harmless manner.
6 Evaluation methods
6.1 Test design
The test design shall meet the requirements of Table 1.

6.5 Removing of heavy metals
6.5.1 Removing of microbiological products
Take 9 pieces of 250 mL triangular flasks, including 3 for blank samples, 3 for
test samples and 3 for deactivated samples. According to the substrate mass
concentration and the volume of addition of microbial products, which are
determined by the optimal reaction system, prepare a culture solution whose
reaction system is 100 mL; perform the removal reaction under the optimal
reaction system conditions.
6.5.2 Removing of enzyme products
Take 9 pieces of 50 mL centrifuge tubes, including 3 for blank samples, 3 for
test samples and 3 for deactivated samples. According to the volume of addition
of enzyme products and the concentration of the substrate, which are
determined by the optimal reaction system, prepare a reaction solution whose
reaction system is 15 mL; perform the removal reaction under the optimal
reaction system conditions.
6.6 Determination
After the removal process, use a centrifuge; continue centrifugation at a speed
of 6 000 r/min for (20±1) min; take the supernatant. Determine the mass
concentration of heavy metals according to the method that is specified in GB/T
5750.6.
6.7 Calculation of heavy metal removal rate
The heavy metal removal rate shall be calculated according to Formula (1):

R – removal rate, %;
ρ0 – the mass concentration of heavy metals in the initial substrate at the
beginning of the reaction, in micrograms per liter (μg/L);
ρ1 – the mass concentration of heavy metals in the residual substrate after the
reaction of the test sample, in micrograms per liter (μg/L);
